Lower White Matter Volume and Worse Executive Functioning Reflected in Higher Levels of Plasma GFAP among Older Adults with and Without Cognitive Impairment

Objective:
There are minimal data directly comparing plasma neurofilament light (NfL) and glial fibrillary acidic protein (GFAP) in aging and neurodegenerative disease research. We evaluated associations of plasma NfL and plasma GFAP with brain volume and cognition in two independent cohorts of older adults diagnosed as clinically normal (CN), mild cognitive impairment (MCI), or Alzheimer’s dementia.
Methods:We studied 121 total participants (Cohort 1: n = 50, age 71.6 ± 6.9 years, 78% CN, 22% MCI; Cohort 2: n = 71, age 72.2 ± 9.2 years, 45% CN, 25% MCI, 30% dementia). Gray and white matter volumes were obtained for total brain and broad subregions of interest (ROIs). Neuropsychological testing evaluated memory, executive functioning, language, and visuospatial abilities. Plasma samples were analyzed in duplicate for NfL and GFAP using single molecule array assays (Quanterix Simoa). Linear regression models with structural MRI and cognitive outcomes included plasma NfL and GFAP simultaneously along with relevant covariates.
Results:Higher plasma GFAP was associated with lower white matter volume in both cohorts for temporal (Cohort 1: β = −0.33, p = .002; Cohort 2: β = −0.36, p = .03) and parietal ROIs (Cohort 1: β = −0.31, p = .01; Cohort 2: β = −0.35, p = .04). No consistent findings emerged for gray matter volumes. Higher plasma GFAP was associated with lower executive function scores (Cohort 1: β = −0.38, p = .01; Cohort 2: β = −0.36, p = .007). Plasma NfL was not associated with gray or white matter volumes, or cognition after adjusting for plasma GFAP.
Conclusions:Plasma GFAP may be more sensitive to white matter and cognitive changes than plasma NfL. Biomarkers reflecting astroglial pathophysiology may capture complex dynamics of aging and neurodegenerative disease.

A series of porous aluminum-based materials are prepared by the reduction of solutions of metal chlorides with lithium powder in diethyl ether under dry argon. The reactants must be combined slowly, but either order of addition is used. The reduction of AlCl3 produces hollow Al balls composed of ~100 nm aluminum particles in nearly quantitative yield after the LiCl byproduct is washed away with dry tetrahydrofuran. Similar structures are formed when mixtures of AlCl3 and SiCl4, are reduced, except that the second component has the effect of reducing the Al nanoparticle size. Mixtures of AlCl3 with FeCl3 reduce to similar ball-like porous structures that are composed of Al, Fe, and Fe-Al intermetallic nanoparticles. When AlCl3 and ZnCl2 are co-reduced, flake-like nanoporous structures are obtained, and solutions of AlCl3 + VCl3 produce more compact nanoporous structures. Some side reactions involving ether cleavage that produces aluminum alkoxides and alkyls do occur, and the amount of side reaction is dependent on the identity of the second metal. The reduction of AlCl3 with excess (4 eq) Li powder produces LiAl nanomaterials. NMR shows the intermetallic compound LiAl to be the only Li-Al intermetallic present.

Solutions or mixtures of aluminum halides AlX3 (X = Cl, Br, I) or aluminum halide alkoxide (ROAlX2; R= i-Pr, X = Cl, Br) are prepared in toluene, or pentamethyldiethylenetriamine (PMDETA) and treated with a slight excess of 50:50 (wt.) sodium potassium alloy (Na/K). Upon agitation, the reaction takes place slowly. The reaction appears to be limited by the deposition of insoluble products on the surface of the Na/K since sonication in an ordinary ultrasonic cleaner is necessary to bring the reaction to completion in several hours. Aluminum nanoparticles (NPs) and Na and K halides are formed, and soluble Al compounds, are also formed in some cases. Lithium powder is used instead of Na/K to reduce Al(N(SiMe3)2)3 to Al NPs. In an effort to passivate the surface of the Al particles to oxidation or hydrolysis, various agents are added either during the reaction or afterward. The products are characterized by SEM, XRD, NMR, and TEM, and the stability to water and air is evaluated.

Synthesis of surface passivated Zr-Al mixed metal-hydride nanoparticles was accomplished via a multi-step process. The initial reaction to produce the zirconium aluminum hydride was via decomposition of zirconium tetrahydroaluminate (Zr(AlH4)4) while exposed to ultrasound produced by a benchtop ultrasonic bath. The particles were surface passivated using carbohydrates and were shown to be stable in air and partially stable in water. TEM imaging suggests the existence of smaller particles made of a Zr-Al alloy that range in size from 1.8 nm to 7.9 nm in diameter and are interspersed with larger particles that range from tens to hundreds of nanometers in diameter. It was also shown that the carbohydrate-derived coating of the nanoparticles is present as an aluminum alkoxide gel surrounding the core particles.
